Cannabinoids, Cannabis, and Oxidative Stress: A Neuroprotective Effect

In the past decade there have been a number of studies that show Cannabis sativa CBD protects neurons from damage or neurodegenerative disorder [6,7], even though its effects in oxidative stress remain unclear. CBD may have neuroprotective effects through restoring a balance between oxidative- and anti-oxidant processes. This ability is often disrupted in many neurodegenerative diseases, as evidenced by increased levels of oxidative damage and reduced antioxidant enzymes. In addition, CBD can also bind to specific receptors and increase the activity of nrf-2/ARE indicating that it may be able to promote neuroprotection by activating these pathways.

In a set of experiments we studied the effects on toxicity of various cannabis extracts and cannabinoids in an in vitro ischemia model for the forebrain affecting the CA1 region of organotypic hippocampal rat slices. The aim of our study was to assess CB1 & CB2 receptors' role in THC & CBD-induced neurotoxicity.


Our results show that the toxicity induced by 1 uM of THC on the CA1 subregion of OGD-exposed slices was completely prevented by CB1 receptor antagonists AM251, but not by CB2 receptor antagonists AM630, and a selective CB1 antagonist WAY-100635, whereas CBD showed a significant neuroprotective effect against OGD-induced cell death in the CA1 subregion of OGD-exposed rat organotypic hippocampal slice only by co-incubation with a TRPV2 antagonist tranilast and a 5HTA1 antagonist WAY-100635 and a PPARg antagonist G3335. The neuroprotective effect 10 uM CBD had was also prevented by the TRPV2 and 5-HT1A receptor antagonists WAY-100635, but not by AM251.
